For me too it's Conker's BFD on N64. I can't remember another one witch make me laugh.
Watching some off my friend playing wii sports is also funny. lol
Conker's BFD, probably followed closely by the Worms saga.

Warioware for wii is pretty funnny.
The Knight Rider PS2 games - they aren't meant to be funny but the gameplay is so daft you can't help but laugh.
Portal was freaking hilarious.
Team Fortress 2 - The whole things like a cartoon. A funny cartoon.
Timesplitters: Future Perfect, its so random and crazy
GTA: SA - The script was so stupid it was funny. I think it was made that way on purpose
Destroy all Humans 1. The second game was funny, too, but not as much as the first game, imo.
The segments with Sheogorath in Oblivion were kind of funny
Disgaea was somewhat funny, but some of the jokes were 5th grade humor, like the horse weiner joke
The Simpsons Game had quite a few laughs in it.

surely some more that I can't recall right now. I think point and click adventure games on the PC contain most of the humour though (see Grim Fandango for example and other excellent titles !)

defianatly should pick this up if you want a laugh. i played it at a friends for half an hour or so and its pretty damn random
